RESUMO

The possible effect of protein-enriched Opuntia cladode supplementation during the pre- and post-partum stages (-25 days to +15 days; day 0 = kidding) upon dam-kid metabolic status, colostrum-milk quality, and some behavioral kid-to-dam interaction in goats managed under rangeland extensive conditions was evaluated. Multiparous crossbred goats (n = 30), homogeneous regarding live weight (LW; 55.9 ± 1.03 kg) and body condition score (BCS; 2.5 ± 0.2 units), were randomly assigned to (1) protein-enriched Opuntia (PEO; n = 10; 29.8% crude protein (CP), 2.2 Mcal ME kg-1), (2) non-enriched Opuntia (NEO; n = 10; 6.4% CP, 2.1 Mcal ME kg-1), and (3) control (CON; n = 10, non-supplemented). The PEO and NEO goats were individually supplemented with Opuntia cladodes (250 g day-1; 09:00-10:00 a.m.; 25 days pre- and 15 days post-partum); then, all groups grazed in a marginal rangeland (10:00 a.m. to 06:00 p.m.). LW, BCS, and blood samples to quantify serum glucose (GLU) levels were collected weekly from day -25 up to day +15 in both dams (pre- and post-partum) and kids (post-partum). At 4h and 8h post-partum, kid-to-dam behavioral tests were performed; approaches (APRO, units), animal-to-animal contact (ACONT, s), latency-to-contact (LCONT, s), and high (HPB) and low (LPB) bleats were registered. The response variables LW (58.2 ± 3.5 kg), GLU from does (66.4 ± 3.3 mg/dL), colostrum fat (12.3 ± 1.15%), non-fatty solids (20.9 ± 2.1%), density (64.4 ± 7.0%), and protein (8.1 ± 0.8%), as well as milk density (31.2 ± 1.7%) and protein (3.9 ± 0.3%), favored the PEO group. Moreover, the dam-to-kid 4 h LPB (34.5 ± 4.6 frequency), as well as kid-to-dam 8 h LCONT-own (100 ± 35.5 s) and LPB (25.2 ± 6.9 frequency) also favored the PEO group. To conclude, peripartum supplementation with protein-enriched Opuntia cladodes emerged as a key alternative to enhance the dam-kid metabolic status, to improve colostrum quality and some milk components (density and protein), as well as to expand the kid-to-dam bond in goat production systems under marginal extensive conditions.

BACKGROUND: Cranberry (Vaccinium macrocarpon L.) fruit quality traits encompass many properties. Although visual appearance and fruit nutritional constitution have usually been the most important attributes, cranberry textural properties such as firmness have recently gained importance in the industry. Fruit firmness has become a quality standard due to the recent demand increase for sweetened and dried cranberries (SDC), which are currently the most profitable cranberry product. Traditionally, this trait has been measured by the cranberry industry using compression tests; however, it is poorly understood how fruit firmness is influenced by other characteristics. RESULTS: In this study, we developed a high-throughput computer-vision method to measure the internal structure of cranberry fruit, which may in turn influence cranberry fruit firmness. We measured the internal structure of 16 cranberry cultivars measured over a 40-day period, representing more than 3000 individual fruit evaluated for 10 different traits. The internal structure data paired with fruit firmness values at each evaluation period allowed us to explore the correlations between firmness and internal morphological characteristics. CONCLUSIONS: Our study highlights the potential use of internal structure and firmness data as a decision-making tool for cranberry processing, especially to determine optimal harvest times and ensure high quality fruit. In particular, this study introduces novel methods to define key parameters of cranberry fruit that have not been characterized in cranberry yet. This project will aid in the future evaluation of cranberry cultivars for in SDC production.

The effect of protein enriched Opuntia cladodes supplementation upon changes of serum total protein, urea, cholesterol, glucose as related to estrus induction (EI%), estrus latency (EL, h), and ovulation rate (OR, units) in adult anestrous goats exposed to the male effect was evaluated. In late April, anestrus goats (n = 45, 25° N) homogeneous regarding live weight (LE; 43.8 ± 1.6 kg) and body condition score (BCS; 2.3 ± 0.1 units) were randomly assigned to: (1). Protein-enriched Opuntia (PEO; n = 15; 29.8% CP, 2.2 Mcal ME kg-1), (2). Non-enriched Opuntia (NEO; n = 15; 6.4% CP, 2.1 Mcal ME kg-1), and (3). Control (CON; n = 15). NEO and PEO goats were individually supplemented with cladodes (160 g d-1; 0900-1000 h), thereafter all groups grazed in a marginal rangeland (1000-1800 h). Neither LW (p > 0.05) nor BCS (p > 0.05) differed among groups, yet an increased (p < 0.05) EI % (100, 57, 42 ± 0.16%), EL h (62, 60, 32 ± 4.2 h), and OR (1.33, 0.71, 0.43 ± 0.23 units) occurred in PEO and NEO vs. CONT, respectively. However, neither blood metabolites differed among groups nor a treatment x time interaction occurred. Peri-breeding protein enriched Opuntia cladodes supplementation of anestrous goats exposed to active males increased (p < 0.01) reproductive outcomes during the non-breeding season.

Immunochemical and mass spectrometric methods were used to examine the gluten composition of a gluten-reduced beer produced by brewing with barley malt in the presence of prolyl endopeptidase (PEP) and a final filtration treatment with diatomaceous earth and perlite. The competitive ELISA is generally considered appropriate for the analysis of hydrolysed gluten, but it is not considered a scientifically valid method for the quantification of gluten in fermented or hydrolysed foods due to the lack of an appropriate reference standard. As no single analytical method can capture the spectrum of gluten-derived products in beer, a comprehensive approach was employed to analyse the intact and hydrolysed fractions of gluten with complementary methods. The combination of PEP addition and diatomaceous earth/perlite filtration was more effective at reducing the concentration of detectable gluten than each of the treatments alone. However, gluten proteins and/or polypeptides were observed in filtered, PEP-treated beers using sandwich ELISA methods, western blot, and bottom-up mass spectrometry. In addition, mass spectrometry results showed that the number of hydrolysed gluten peptides was almost unaffected by the filtration process. Gluten peptides that contained potentially immunopathogenic sequences were identified in the filtered PEP-containing beers by MS. Variability in gluten composition was observed between three replicate pilot-scale productions, suggesting that the gluten profile in beer could differ from batch to batch. As there is uncertainty in the detection and quantification of gluten in hydrolysed and fermented foods, characterisation of hydrolysed gluten by complementary analytical methodologies is recommended.

Introducción: La expresión variable en la sintomatología de los trastornos temporomandibulares (TTM) es un desafío para el clínico; así, el origen del dolor y la estructura comprometida no son fáciles de identificar. El disco articular está directamente vinculado a la función mandibular y, en condiciones patológicas, puede verse alterada su posición y consecuentemente su zona de carga. Si bien la presencia de terminaciones nerviosas y los troncos nerviosos potencialmente involucrados en su inervación han sido identificados, la forma en que los nervios se distribuyen al interior del disco no se ha precisado aún. El objetivo de este estudio fue describir la distribución nerviosa del disco articular de la articulación temporomandibular en cadáveres humanos. Material y método: Se realizó disección anatómica de ocho discos articulares de articulaciones temporomandibulares del lado derecho de cadáveres conservados. Los discos articulares fueron sometidos al método de tinción de Sihler y observados bajo lupa estereoscópica y transiluminación. Se realizó fotografía estandarizada y se dividió el disco en 5 zonas (medial, lateral, anterior, posterior, central) caracterizando la distribución nerviosa. Resultados: Se observó presencia de estructuras nerviosas en todas las zonas excepto en la zona central. Las estructuras nerviosas discurren como troncos simples o múltiples en dirección antero-posterior con un número variable de colaterales en las áreas lateral y medial del disco, comunicando la zona retrodiscal con la porción posterior del músculo pterigoideo lateral en todas las muestras. Conclusión: Existe un patrón de inervación común en los discos estudiados. El disco se encuentra inervado en su periferia por troncos nerviosos que presentan un trayecto anteroposterior. La comunicación entre distintas regiones anatómicas por continuidad nerviosa, así como el patrón de inervación del disco, podrían explicar situaciones de dolor local y referido en TTM


Introduction: Variable expression of symptomatology in temporomandibular disorders (TMD) is a challenge for clinician, because source of pain is not easy to identify. Articular disc is directly linked to the mandibular function and in pathological conditions can be altered its position and consequelty its zone of load. The aim of this study was to describe the intradiscal nerve distribution of temporomandibular joint disc in human cadavers. Although the presence of sensitive nerve terminals in the disc and the nerve trunks potentially involved have been identified, the way the nerves are distributed inside disc remains unprecised. Material and method: Anatomical dissection of 8 articular discs of the temporomandibular joints on the right side of preserved cadavers was performed. The articular discs were subjected to the Sihler staining method and then observed under a stereoscopic loupe and transillumination, A standardized photograph was perform by arbitrarily dividing the disk into 5 zones (medial, lateral, anterior, posterior, central) and characterized thebnerve distribution. Results: Presence of nerve structures was observed in all areas except the central zone. Nerve structures run as a single or multiple trunk in anterior-posterior direction with a variable number of collateral in lateral and medial areas of disc, comunicating retrodiscal zone and posterior zone of lateral pterigoid muscle in all samples. Conclusion: It can be identified a common inervation pattern in all discs. The disc is innervated in its periphery by nerve trunks that present an anterioriposterior path. The communication between different anatomical regions by nervous continuity, as well as the innervation pattern of the disc, could explain situations of local pain and referred in TMD

The possible influence of the "male effect" upon reproductive outcomes of adult anestrous goats under marginal rangeland conditions and supplemented with protein-enriched Opuntia megacantha Salm-Dyck was evaluated. Reproductive variables included: estrus percentage (EST, %), estrus latency (ESL, hours), ovulation percentage (OP, %), ovulation rate (OR, units), average largest follicle at ovulation (LFO, mm), largest corpus luteum (LCL, mm), embryo number (EBN, units), and embryo implantation percentage (EIP, %). During early May, anestrous mix-breed adult goats (Criollo x Alpine-Saanen-Nubian; n = 38, 26° N) were randomly distributed to (1) Control (CC; n = 12), (2), Non-enriched Opuntia (NEO; n = 14), and (3) Protein-enriched Opuntia (PEO; n = 12). Neither LW (P > 0.05) nor BCS (P > 0.05) or any of the evaluated ovarian variables differed (P > 0.05) among treatments; EST = 89.66%, ESL = 53.66 h, OP = 70.33%, OR = 1.07 units, LFO = 4.5 mm, LCL = 9.6 mm, EBN = 0.94 embryos, and EIP = 48.66%. Irrespective of nutritional supplementation regime, all goats denoted an increased response to the male effect just in the middle of the anestrous season and managed under marginal grazing conditions during the dry season (May to June; 26° N). The use of the male effect successfully invoked neurophysiological pathways to re-activate ovarian follicular and luteal pathways during the natural anestrous season in the female goat. Yet, such successful physiological scenario was not equally exerted to promote an increased embryo implantation rate; this issue claims further consideration. Therefore, it is essential to align not only the peri-conceptional but also the peri-implantation stages to the best suited environmental conditions in the rangeland, in order to increase both reproductive and economic efficiency while promoting sustainability in those rangeland-based marginal goat production systems.

An immobilized lipase from Thermomyces lanuginosus (TL IM) was employed to mediate the continuous transesterification of sesame oil and fully hydrogenated soybean oil (FHSBO) in a packed-bed reactor operating at 70 degrees C. Reactions between sesame oil (rich in LLL (15.97%), LOL (31.56%), and OLO (21.15%) [L = linoleic; O = oleic]) and the fully hydrogenated fat ((73.7% SSS, 26.3% SPS) [S = stearic; P = palmitic]) produced semi-solid fats. These products are complex mixtures of triacylglycerol (TAG) species whose compositions depend on reaction conditions. The dependence of the steady state product TAG profile on space time was determined for four initial weight ratios of sesame oil to hydrogenated fat (90:10, 80:20, 70:30, and 60:40). Except for the trial involving a weight ratio of sesame oil to FHSBO of 60:40, near equilibrium conditions were achieved at space times of 30 min-1 h. The chemical, physical, and functional properties of the product semi-solid fats were characterized. The predominant TAG species in the quasi-equilibrium products obtained from the mixture initially containing 90% (w/w) sesame oil and 10% FHSBO were LOL (26.22%) and OLO (21.92%). For transesterification of 80% sesame oil and 20% FHSBO, the major product species were OOP (21.27%), LOL (17.46%), and OLO (13.93%). OOP (24.38%) was the major product for reaction of 70% sesame oil with 30% FHSBO. Appropriate choices of reaction conditions and initial ratios of sesame oil to FHSBO lead to TAG with melting profiles and solid fat contents (SFC) similar to those of a variety of commercial products.
